How does Vardamate 20 Tablet work?

Vardenafil is a phosphodiesterase (PDE-5) inhibitor. It works in erectile dysfunction by increasing blood flow to the penis by relaxing the muscles in penile blood vessels. It helps to get an erection only if the person is sexually stimulated.

• Role of PDE5 Inhibition: By inhibiting PDE5, vardenafil increases cGMP levels, thereby improving erectile function. However, sexual stimulation is necessary for the local release of nitric oxide, meaning PDE5 inhibition alone does not induce an erection without sexual arousal.

Dosage & Administration:

Recommended Starting Dose:

• Standard Dose: 10 mg once daily, taken orally about 60 minutes before sexual activity.

Interaction

Antihypertensive Drugs:

• Caution: Advised when co-administered with other vasodilators, including alpha-blockers.

CYP3A4 Inhibitors:

• Examples: Ketoconazole, indinavir, ritonavir, and erythromycin.
• Effect: These inhibitors may decrease the excretion of vardenafil from the body, potentially increasing its effects and side effects.

Contraindications

• Vardenafil can potentiate the hypotensive effect of nitrates.

Pregnancy & Lactation

Vardenafil is not indicated for women.

Precautions & Warnings

Physicians should consider the cardiovascular status of their patients, as there is a certain degree of cardiac risk associated with sexual activity. In men who are discouraged from being sexually active due to their underlying cardiovascular condition, vardenafil should generally not be used.
